Desmume has, if I remember correctly, 2 or 3 different audio backends. Try those and if they don't work, it may help to come back with specs and settings used. Sometimes changing the audio thing used can help, but it won't always.
I downloaded Spirit Tracks earlier today to try out, and I've just reached the cutscene in the castle where we meet princess Zelda for the first time and when Zelda's Lullaby started the music suddenly became super staticky and distorted, like it was peaking or something. I haven't encountered this on any of the other ROMs I've played (mainly pokemon) and the sound quality was fine up till that point. I'm using the DeSmuMe emulator. Is this an issue with the game file itself or are there some emulation settings I can tweak to fix it?
Update I was about to post this and decided to try speeding through the cutscene and the sound fixed itself after, so it's probably just the one song file that's messed up.
